Mikroskobik gölge görüntüleme tabanlı mikroakışkan sitometrisi
Özet
Hücre sayımı ve konsantrasyonlarının belirlenmesi teşhiste, tedavinin takibinde ve bilimsel araştırmalarda sıklıkla kullanılmaktadır. Akış sitometrisi hücreleri otomatik olarak sayan ve konsantrasyonlarını belirleyen standart bir yöntemdir. Araştırma amaçlı ve klinik teşhiste yaygın olarak kullanılmaktadır. Klasik akış sitometrisi hücreleri görüntülemeden, ışık saçılımı, geçişi ve emilimi ilkelerine dayanarak sayar. Cihaz büyük, hantal ve pahalıdır ayrıca oldukça gelişmiş bir optik ölçüm yöntemi olması nedeniyle yerinde teşhis uygulamaları için uygun değildir. Erken teşhis ve tedavinin hızlanması için yerinde teşhis önemlidir. Ayrıca kaynak kısıtlı bölgelerde gelişmiş laboratuvarlar olamayabileceği için yerinde teşhis yapabilen cihazlara ihtiyaç duyulmaktadır. Hücrelerin üsten aydınlatılmasıyla gölge görüntülerinin bir mikro akışkan içinde elde edilmesine mikroskobik gölge görüntüleme denir. Bu yöntem geniş alan lensiz bir görüntüleme sistemidir. Bu tez çalışmasında akış sitometirisi kanalına benzer bir mikro kanal düzeneği ve hücreler akış halindeyken mikro kanal içinde gölge görüntüleme yöntemiyle görüntüleyen bir geniş alan mikroskop sistemi tasarlanmıştır ve üretilmiştir. Elde edilen görüntülerden otomatik hücre sayımını gerçekleştiren Hough Dönüşümü tekniğine dayanan bir yazılım da üretilmiştir. Sistem küçük ve performansı düşük bir masa üstü bilgisayarla rahatlıkla çalışmaktadır, boyutları 15cmx15cmx30cm’dir. Tüm tasarım yaklaşık 1500 Avro’ya mal olmuştur, prototip için kullanılan gelişmiş kamera ve ışık sistemi için uygun maliyetli çözümler önerildiğinde 200 Avro’ya kadar maliyet azaltılabilir. Sistem 40 ul solüsyondaki 1666 hücreyi 1 saniyede sayabilmektedir. Sistem %87,75 hassaslık oranına ve %77,77 özgüllük oranına sahiptir. Akış sitometrisinin yerinde teşhiste kullanılabilmesi toplumsal ve ekonomik yarar getirebilir.Determination of number of cells and concentrations are frequently used in diagnosis,

automatically counts cells and determines their concentrations. It is widely used for research
purposes and clinical diagnosis. Classical flow cytometry counts cells without imaging them,
it’s counting idea is based on the principles of light scattering, transmission, and absorption.
The device is large, bulky and expensive, and is not suitable for point of care diagnostic
applications due to its highly advanced optical measurement method. Point of care diagnosis
is important for early diagnosis and faster treatment. In addition, since there may not be
developed laboratories in resource-restricted settlements, devices that can make point of care
diagnosis are needed. Obtaining shadow images in a microfluidic channel by illuminating
the cells from top is called microscopic shadow imaging. This method is a wide field lensless
imaging system.
In this thesis, a micro-channel setup similar to flow cytometry channel and a wide-field
microscope system that displays cells in the micro-channel with shadow imaging method
was designed and produced. A software based on the Hough Transform technique that
performs automatic cell counting from the images obtained was also produced. The system
works practically with a small and low performance laptop computer, the decive dimensions
are 15cmx15cmx30cm. The entire design cost about 1500 Euros, when cost-effective
solutions are offered for the advanced camera and light system used for the prototype, the
cost can be reduced to 200 Euros. The system can count 1666 cells in 40 ul of solution in a
second. The system has 87.75% sensitivity and 77.77% specificity. The ability to use flow
cytometry in point of care diagnosis can bring social and economic benefits.
