Taşınabilir ve düşük maliyetli kuvars akort çatal sürücü tabanlı prototip üretimi ve immunosensör olarak yapılandırılması
Özet
Canlılar, çevrede meydana gelen değişiklikleri duyu organları yoluyla algılayarak hayatlarını bu değişimlere göre şekillendirmektedir. Doğayı taklit ederek çeşitli hastalıkların teşhisinde biyosensör yapılandırılma stratejilerinin kullanımı, tıpkı duyu organları gibi ortamdaki değişikleri anlamlandırma yaklaşımıdır. Bu tez çalışmasında öncelikle, birinci nesil olarak adlandırılan bununla birlikte patent değeri olan kuvars akort çatalı (QTF) sürücüsünü tanımlamak ve 118S614 numaralı TÜBİTAK destekli proje kapsamında ise bu cihazın geliştirilmesiyle ikinci nesil olarak adlandırılan cihazı tanıtmaktır. Tezin ana kapsamı ise, seçilen iki farklı analitin (transferrin ve kanda serbest dolaşan tümör hücresi) yeni bir yaklaşım olan kuvars akort çatalı temelli kütle hassas immünosensör yapılandırılması ile teşhis edilebilir olup olmadığını araştırmaktır. Bu iki analit özel olarak seçilmiştir. Bu seçimin nedeni ise, iki analitten ilki olan transferrin, hastalık öyküsünün sıkça grip nezle ile karıştırıldığı ve görüntüleme yöntemlerinin yetersiz kaldığı durumlarda beyin omurilik sıvı kaçaklarına tanı koymada önemli bir biyoişaret molekülü olmasıdır. Öte yandan seçilen diğer analit olan kanda serbest dolaşan tümör hücresi ise kanserin erken evrede teşhis edilebilmesini sağlayan oldukça önemli bir biyoişaret molekülüdür. Bahsedilen amaçlar doğrultusunda QTF kütle hassas immünosensörü yapılandırılmış ve önemli veriler elde edilmiştir (R2=0,9686).
Living things perceive changes in the environment through their sensory organs and shape their lives according to these changes. The use of biosensor structuring strategies in the diagnosis of various diseases by imitating nature is an approach to make sense of changes in the environment, just like the sense organs. In this thesis, first of all, it is to define the quartz tuning fork (QTF) driver, which is called the first generation and has a patent value, and to introduce the device called the second generation with the development of this device within the scope of the TÜBİTAK supported project numbered 118S614. The main scope of the thesis is to investigate whether two selected analytes (transferrin and circulating tumor cell in blood) can be diagnosed with a new approach, a quartz tuning fork-based mass-sensitive immunosensor configuration. These two analytes are specially selected. The reason for this choice is that transferrin, the first of the two analytes, is an important biomarker molecule in diagnosing cerebrospinal fluid leaks in cases where the history of the disease is frequently confused with influenza and imaging methods are insufficient. On the other hand, the other analyte selected, the circulating tumor cell in the blood, is a very important biomarker molecule that enables cancer to be diagnosed at an early stage. QTF mass sensitive immunosensor was configured for the aforementioned purposes and important data were obtained (R2=0,9686).
